分泌及び膜貫通ポリペプチドとそれをコードしている核酸

【課題】ヒトに於ける、新規細胞外タンパク質の存在及び機能の同定及び上記タンパク質をコードする核酸分子配列を提供する。
【解決手段】新規ポリペプチド及びこれらペプチドをコードする核酸分子を対象としている。また、ここにおいて提供されるのは、これら核酸配列を含んでなるベクター及び宿主細胞、異種ポリペプチド配列に融合したポリペプチドを含んでなるキメラポリペプチド分子、ポリペプチドと結合する抗体、並びにポリペプチドを製造する。

【特許請求の範囲】
【請求項１】
以下の（a）又は（b）のヌクレオチド配列を含んでなる単離された核酸：
（a）配列番号：５８に示すアミノ酸をコードするヌクレオチド配列、及び
（b）配列番号：５８に示すアミノ酸配列において、1若しくは数個のアミノ酸が欠失、置換若しくは付加された、配列番号：５８に示すアミノ酸配列から由来するポリペプチドをコードするヌクレオチド配列であって、かつ前記ヌクレオチド配列からなる核酸が大腸癌に過剰発現するヌクレオチド配列。
【請求項２】
以下の（a）又は（b）のヌクレオチド配列を含んでなる単離された核酸：
（a）配列番号：５７に示されるヌクレオチド配列、及び
（b）配列番号：５７に示されるヌクレオチド配列の核酸に対し相補的なヌクレオチド配列からなる核酸と高度の緊縮性条件下でハイブリダイズするヌクレオチド配列であって、かつ前記ヌクレオチド配列からなる核酸が大腸癌に過剰発現するヌクレオチド配列。

以下の（a）又は（b）のヌクレオチド配列を含んでなる単離された核酸。
（a）配列番号：５７に示されるヌクレオチド配列の全長コード配列、及び
（b）配列番号：５７に示されるヌクレオチド配列の核酸に対し相補的なヌクレオチド配列からなる核酸と高度の緊縮性条件下でハイブリダイズするヌクレオチド配列であって、かつ前記ヌクレオチド配列からなる核酸が大腸癌に過剰発現するヌクレオチド配列。

請求項１ないし３のいずれか１項に記載の核酸に対し相補的なヌクレオチド配列を含んでなる単離された核酸。
【請求項５】
請求項１ないし４のいずれか１項に記載の核酸を含んでなるベクター。
【請求項６】
ベクターで形質転換された宿主細胞によって認識されるコントロール配列と作用可能に結合した請求項５に記載のベクター。
【請求項７】
請求項５又は６に記載のベクターを含んでなる宿主細胞。
【請求項８】
前記細胞がCHO細胞である、請求項７に記載の宿主細胞。
【請求項９】
前記細胞が大腸菌である、請求項７に記載の宿主細胞。
【請求項１０】
前記細胞が酵母細胞である、請求項７に記載の宿主細胞。
【請求項１１】
請求項８ないし１０のいずれか１項に記載の宿主細胞をポリペプチドの発現に適した条件下で培養し、細胞培養物から前記ポリペプチドを回収することを含んでなる、前記ポリペプチドの製造方法。
【請求項１２】
以下の（a）又は（b）のアミノ酸配列を含む単離された天然配列ポリペプチド：
（a）配列番号：５８に示すアミノ酸配列、及び
（b）アミノ酸配列（a）において1若しくは数個のアミノ酸が欠失、置換若しくは付加された、（a）から由来するアミノ酸配列であって、かつ前記アミノ酸配列からなるポリペプチドをコードする核酸が大腸癌に過剰発現するアミノ酸配列。

異種アミノ酸配列に融合した、請求項１２に記載のポリペプチドを含んでなるキメラ分子。
【請求項１４】
前記異種アミノ酸配列がエピトープダグ配列である、請求項１３に記載のキメラ分子。
【請求項１５】
前記異種アミノ酸配列が免疫グロブリンのFc領域である、請求項１３に記載のキメラ分子。
【請求項１６】
請求項１２に記載のポリペプチドへ特異的に結合する抗体。
【請求項１７】
前記抗体がモノクローナル抗体、ヒト化抗体、ヒト抗体または単鎖抗体である、請求項１６に記載の抗体。
【請求項１８】
哺乳類被験者の身体から分離された試験生物学的試料中の配列番号：５８のポリペプチドをコードする核酸のレベルを、対応する正常生物学的試料中の前記核酸のレベルと比較して決定することを含んでなる、前記試験生物学的試料中の前記核酸のレベルの増加が試験生物学的試料中に癌細胞を含むことを示す診断方法。

試験生物学的試料または正常生物学的試料が組織試料である、請求項１８に記載の方法。
【請求項２０】
試験組織試料を大腸組織から採取する、請求項１９に記載の方法。
【請求項２１】
試験生物学的試料又は正常生物学的試料から得られた核酸のレベルを、配列番号：５８のポリペプチドをコードする核酸に特異的なプローブを１つもしくはそれ以上用いてハイブリダイゼーションすることにより決定する請求項１８に記載の方法。
【請求項２２】
ハイブリダイゼーションが高度の緊縮性条件下で行われる、請求項２１に記載の方法。
【請求項２３】
試験生物学的試料又は正常生物学的試料から得られた核酸がcDNAである、請求項２１に記載の方法。
【請求項２４】
試験生物学的試料又は正常生物学的試料から得られた核酸がマイクロアレイに置かれる、請求項２３に記載の方法。
【請求項２５】
正常生物学的試料と比較して、哺乳類被験者の身体から分離された試験生物学的試料中の配列番号：５８のポリペプチドの発現レベルを決定することを含んでなる、前記試験生物学的試料中の前記ポリペプチドの過剰発現が試験生物学的試料中に癌細胞を含むことを示す診断方法。

配列番号：５８のポリペプチドに特異的に結合する抗体を用いて過剰発現を検出する請求項２５に記載の方法。
【請求項２７】
前記抗体がモノクローナル抗体、ヒト抗体、ヒト化抗体または単鎖抗体である、請求項２６に記載の方法。

前記抗体が抗体断片である、請求項２６に記載の方法。
