イソプレンと副生成物の産出方法

【特許請求の範囲】
【請求項１】
イソプレンシンターゼポリペプチドをコードする異種核酸を含む、酸素制限条件下で、イソプレンと、エタノール、１，３−プロパンジオール、および水素からなる群から選択される副生成物とを同時に生成することができる細胞であって、前記細胞が（ｉ）約０．１ｍｇ／Ｌ液体培地／時間を上回るイソプレンの平均容積生産性と、約０．１ｍｇ／Ｌ液体培地／時間を上回る副生成物の平均容積生産性を有するか；または（ｉｉ）約４００ｎｍｏｌｅ／ｇｗｃｍ／時間〜約２．０×１０５ｎｍｏｌｅ／ｇｗｃｍ／時間の速度でイソプレンを産出し、約０．０１ｍｍｏｌ／Ｌ液体培地／時間〜約２００ｍｍｏｌ／Ｌ液体培地／時間の速度で副生成物を産出する、細胞。
【請求項２】
前記細胞を酸素制限培養で増殖させる、請求項１記載の細胞。
【請求項３】
前記イソプレンシンターゼポリペプチドをコードする異種核酸がプロモーターに機能的に連結されている、請求項１記載の細胞。
【請求項４】
前記イソプレンシンターゼポリペプチドが植物イソプレンシンターゼポリペプチドである、請求項３記載の細胞。
【請求項５】
前記植物イソプレンシンターゼポリペプチドが、ウラジロハコヤナギ（Ｐｏｐｕｌｕｓ ａｌｂａ）由来のものである、請求項４記載の細胞。
【請求項６】
ＭＶＡ経路ポリペプチド、ＤＸＳポリペプチド、またはＩＤＩポリペプチドをコードする異種核酸をさらに含む、請求項５記載の細胞。
【請求項７】
前記副生成物がエタノールである、請求項１記載の細胞。
【請求項８】
エタノール発酵に関与するポリペプチドをコードする異種核酸をさらに含む、請求項７記載の細胞。
【請求項９】
前記エタノール発酵に関与するポリペプチドが、アルコールデヒドロゲナーゼＢ（ａｄｈＢ）ポリペプチド、アルコールデヒドロゲナーゼＥ（ａｄｈＥ）ポリペプチド、またはピルビン酸デカルボキシラーゼ（ｐｄｃ）ポリペプチドである、請求項８記載の細胞。

前記副生成物が１，３−プロパンジオールである、請求項１記載の細胞。
【請求項１１】
グリセロール経路または１，３−プロパンジオール経路に関与するポリペプチドをコードする異種核酸をさらに含む、請求項１０記載の細胞。

前記グリセロール経路または１，３−プロパンジオール経路に関与するポリペプチドが、ジヒドロキシアセトンリン酸レダクターゼ（ＤＡＲ１）、グリセロール−リン酸ホスファターゼ（ＧＰＰ２）、グリセロールデヒドラターゼＢ１（ｄｈａＢ１）、グリセロールデヒドラターゼＢ２（ｄｈａＢ２）、グリセロールデヒドラターゼＢ３（ｄｈａＢ３）、ｄｈａＸ、ｏｒｆＸ、ｏｒｆＹ、１，３−プロパンジオールオキシドレダクターゼ（ｄｈａＴ）、グリセロールデヒドロゲナーゼ（ｄｈａＤ）、またはジヒドロキシアセトンキナーゼ（ｄｈａＫ）である、請求項１１記載の細胞。

前記副生成物が水素である、請求項１記載の細胞。
【請求項１４】
ヒドロゲナーゼポリペプチドをコードする異種核酸をさらに含む、請求項１３記載の細胞。
【請求項１５】
前記ヒドロゲナーゼポリペプチドが、フェレドキシン依存的ヒドロゲナーゼポリペプチド、ＮＡＤＰＨ依存的ヒドロゲナーゼポリペプチド、または酸素耐性ヒドロゲナーゼポリペプチドである、請求項１４記載の細胞。
【請求項１６】
イソプレンと副生成物を同時に生成する方法であって、
（ａ）イソプレンと副生成物の同時生成に好適な条件下で、イソプレンと、エタノール、１，３−プロパンジオール、および水素からなる群から選択される副生成物とを同時に生成することができる細胞を培養すること、ここで、前記細胞は、イソプレンシンターゼポリペプチドをコードする異種核酸を含むこと、と
（ｂ）イソプレンと前記副生成物を同時に生成すること、ここで、前記細胞は（ｉ）約０．１ｍｇ／Ｌ液体培地／時間を上回るイソプレンの平均容積生産性と、約０．１ｍｇ／Ｌ液体培地／時間を上回る副生成物の平均容積生産性を有するか；または（ｉｉ）約４００ｎｍｏｌｅ／ｇｗｃｍ／時間〜約２．０×１０５ｎｍｏｌｅ／ｇｗｃｍ／時間の速度でイソプレンを産出し、約０．０１ｍｍｏｌ／Ｌ液体培地／時間〜約２００ｍｍｏｌ／Ｌ液体培地／時間の速度で副生成物を産出すること、を含む方法。
【請求項１７】
前記細胞を酸素制限培養で増殖させる、請求項１６記載の方法。
【請求項１８】
前記イソプレンシンターゼポリペプチドをコードする異種核酸がプロモーターに機能的に連結されている、請求項１６記載の方法。
【請求項１９】
前記イソプレンシンターゼポリペプチドが植物イソプレンシンターゼポリペプチドである、請求項１８記載の方法。
【請求項２０】
前記植物イソプレンシンターゼポリペプチドが、ウラジロハコヤナギ由来のものである、請求項１９記載の方法。

前記細胞が、ＭＶＡ経路ポリペプチド、ＤＸＳポリペプチド、またはＩＤＩポリペプチドをコードする異種核酸をさらに含む、請求項２０記載の方法。

前記副生成物がエタノールである、請求項１６記載の方法。

前記細胞が、エタノール発酵に関与するポリペプチドをコードする異種核酸をさらに含む、請求項２２記載の方法。
