医薬使用のためのリパーゼ変異体

任意にはプロテアーゼ及び/又はアミラーゼと組合しての、配列番号2のアミノ酸1-269を含んで成るサーモミセス・ラヌギノサス(Thermomyces lanuginosus)(ヒューミコラ・ラヌギノサ(Humicola lanuginose))リパーゼに関連するリパーゼの医薬使用に関する。医学的適応の例は、消化異常、膵臓の外分泌不十分(PEI)、膵炎、嚢胞性線維症、I型及びII型糖尿病の処理である。本発明のリパーゼは、例えばインビトロでの改良された消化性能、中性範囲のpHでの改良された活性、低いpHでの改良された安定性、プロテアーゼ分解に対する安定性、及び/又はペプシン及び胆汁塩の存在下での安定性を有する。本発明はまた、リパーゼのインビトロでの消化性能を決定する方法、及びT. ラヌギノサスのリパーゼの一定の新規変異体にも関する。

【請求項1】
(a)配列番号2のアミノ酸1〜269の配列に対して少なくとも50%の同一性を有し;
(b)リパーゼ活性を有し;そして
(c)配列番号2のアミノ酸1〜269の配列に比較して、置換N33Q、T231R及びN233R、並びに下記:

【請求項2】
次の置換:N33Q+T231R+N233Rを有する、配列番号2の配列を有する対照リパーゼに比較して、インビトロで改良された消化性能を有する請求項1記載のリパーゼ。
【請求項3】
a)対照リパーゼを選択し;
b)それぞれ、100部/体積の食餌と、20部/体積のペプシン及び30部/体積のリパーゼ又は対照リパーゼとを混合し;
c)i)0又はii)10部/体積の緩衝液(0.8MのMES、0.8Mの酢酸ナトリウム、0.8Mのイミダゾール、pH7.0)を添加し、ここで段階i)はpH3の胃段階として言及され得、そして段階ii)はpH5の胃段階として言及され得;
d)振盪下で37℃で1時間インキュベートし;
e)20部/体積の胆汁塩、及びi)25又はii)15部/体積の緩衝液(0.8MのMES、0.8Mの酢酸ナトリウム、0.8Mのイミダゾール、pH7.0)を添加し、ここで段階i)はpH3の胃段階として言及され得、そして段階ii)はpH5の胃段階として言及され得;
f)振盪下で37℃で2時間インキュベートし;

【請求項4】
a)i)対照リパーゼを選択し;
ii)それぞれ、5部/体積のリパーゼ又は対照リパーゼと、5部/体積の
a)0.001%のTriton−X100及び10mMのNaClを含む希釈剤、又は
b)150μg/mlのペプシン、4mMのCaCl2、0.01%のTriton-X100及び50mMのシトレート、pH3.0を含むペプシン処理溶液(ここでa)は未処理のサンプルとして言及され、そしてb)はペプシン処理されたサンプルとして言及される)とを混合し;
iii)段階ii)のサンプルを、20℃で3時間インキュベートし;
iv)段階iii)の個々のサンプルに、1mMのPNP−パルミテート、1.2%のTriton-X100、4mMのCaCl2、100mMのTRIS, pH8.0を含む、55部/体積の基質、及び
a)5部/体積のペプシン処理溶液、又は
b)5部/体積の希釈剤(ここでa)は未処理のサンプルを言及し、そしてb)はペプシン処理されたサンプルを言及する)を添加し;
v)iv)サンプルのOD405を一定間隔で読み取ることにより基質の分解を追跡し;
vi)直線範囲に落ちるv)からのデータを集め、そしてそれぞれ、ペプシン処理されたサンプル及び未処理のサンプルについて、mOD(ミリOD)/時でのリパーゼ活性を計算し;
vii)ペプシン処理されたサンプルのリパーゼ活性を、未処理のサンプルのリパーゼ活性により、それらが段階vi)に起因する場合、割算し、そしてその結果に100を掛算することにより%残留リパーゼ活性(%RA)を計算し;そして所望には、
viii)リパーゼの%RAと、対照リパーゼの%RAとを比較することにより、ペプシンの存在下でpH3での安定性を決定し、そして/又は
b)i)対照サンプルを選択し;
ii)それぞれ、10部/体積のリパーゼ又は対照リパーゼと、23部/体積のa)水、又はb)20mMの胆汁塩とを混合し、ここでa)は未処理のサンプルとして言及され、そしてb)は胆汁塩サンプルとして言及され;
iii)ii)の個々のサンプルに、25mMのスクシネート中、1mMのPNPオレエート、2mMのCaCl2、1.2%のTriton−X100、pH5.0を含む、200部/体積の基質を転化し;
iv)段階iii)の直後、個々のサンプルから、60部/体積の前記得られる混合物を除き、そしてその15部/体積を、4つの別々の区画中に4度、移し;
v)1,2,3及び4時間後、60部/体積の100mMのTRIS(pH8.0)を、iv)の4つの区画のそれぞれの区画に添加し、すぐにOD405を読み取り、そして1,2,3及び4時間の読み取りの線状範囲に基づいて、mOD/時での活性を計算し;
vi)それぞれ、リパーゼ及び対照リパーゼの胆汁塩安定性比率に達するよう、リパーゼ及び対照リパーゼについて、胆汁塩サンプルの段階v)で得られる活性を、段階v)でまた得られるような未処理のサンプルの活性により割算し;そして
vii)リパーゼの胆汁塩安定性比率を、対照リパーゼの胆汁塩を安定性比率により割算する(得られる比率はリパーゼの改良因子として定義され得る)ことにより、胆汁塩の存在下でpH5での活性を定義する段階を含んで成る、対照リパーゼに比較して、リパーゼのインビトロでの消化性能を決定するための方法。

前記インビトロでの消化性能が請求項3又は4のいずれか1項記載の方法を用いて決定される請求項2記載のリパーゼ。

薬剤として使用のためにプロテアーゼ及びアミラーゼと組合しての請求項1〜2及び5〜6のいずれか1項記載のリパーゼ。
【請求項12】
(i)前記プロテアーゼが、
a)配列番号3のアミノ酸1〜274の配列を有するプロテアーゼ、
b)配列番号4のアミノ酸1〜188の配列を有するプロテアーゼ、及び
c)配列番号5のアミノ酸1〜188の配列を有するプロテアーゼから成る群から選択されたプロテアーゼに対して少なくとも70%の同一性を有し;
(ii)前記アミラーゼが、
a)配列番号6のアミノ酸1〜481の配列を有するアミラーゼ、
b)配列番号7のアミノ酸1〜481の配列を有するアミラーゼ、及び
c)配列番号8のアミノ酸1〜483の配列を有するアミラーゼから成る群から選択されたアミラーゼに対して少なくとも70%の同一性を有する請求項10又は11記載のプロテアーゼ及び/又はアミラーゼと組合してのリパーゼ。
【請求項13】
消化障害、膵臓外分泌機能不全、膵炎、嚢胞性線維症、糖尿病I型及び/又は糖尿病II型の処理のための薬剤の製造のためへの請求項1〜2及び5〜6のいずれか1項記載のリパーゼ又はリパーゼ混合物の使用。

プロテアーゼ又はアミラーゼの使用をさらに含んで成る請求項13記載の使用。
【請求項15】
プロテアーゼ及びアミラーゼの使用をさらに含んで成る請求項13記載の使用。
【請求項16】
前記プロテアーゼ及び/又はアミラーゼが、請求項12に記載の通りである請求項14又は15記載の使用。

消化障害、膵臓外分泌機能不全、膵炎、嚢胞性線維症、糖尿病I型及び/又は糖尿病II型の処理への使用のためへの請求項1〜2及び5〜6のいずれか1項記載のリパーゼ。

プロテアーゼ又はアミラーゼと組合しての請求項17記載のリパーゼ。

プロテアーゼ及びアミラーゼと組合して請求項17記載のリパーゼ。

前記プロテアーゼ及び/又はアミラーゼが請求項12記載の通りである請求項18又は19記載のリパーゼ。

請求項1〜2及び5〜6のいずれか1項記載のリパーゼ又はリパーゼ混合物、及び少なくとも1つの医薬的に許容できる助剤材料を含んで成る医薬組成物。

プロテアーゼ又はアミラーゼをさらに含んで成る請求項21記載の組成物。
【請求項23】
プロテアーゼ及びアミラーゼをさらに含んで成る請求項21記載の組成物。
【請求項24】
前記プロテアーゼ及び/又はアミラーゼが請求項12記載の通りである請求項22又は23記載の組成物。
【請求項25】
治療的有効量の請求項1〜2及び5〜6のいずれか1項記載のリパーゼ又はリパーゼ混合物を投与することによる、消化障害、膵臓外分泌機能不全、膵炎、嚢胞性線維症、糖尿病I型及び/又は糖尿病II型の処理方法。

治療有効量のプロテアーゼ又はアミラーゼを投与することをさらに含んで成る請求項25記載の方法。

治療有効量のプロテアーゼ及びアミラーゼを投与することをさらに含んで成る請求項25記載の方法。

前記プロテアーゼ及び/又はアミラーゼが請求項12記載の通りである請求項26又は27記載の方法。

(b)リパーゼ活性を有し;そして
(c)位置30、42、114及び/又は163の少なくとも1つの位置で少なくとも1つの置換を含んで成る(ここで、個々の位置は配列番号2のアミノ酸1〜269の位置に対応する)、親リパーゼの変異体である、薬剤として使用するためのリパーゼ。

(a)配列番号2のアミノ酸1〜269に対して少なくとも50%の同一性を有し;そして
(b)リパーゼ活性を有し;そして
(c)次の置換:A30T、A30V、P42L、T114I及びG163Kから選択された少なくとも1つの置換を含んで成る(ここで、個々の位置は配列番号2のアミノ酸1〜269の位置に対応する)、親リパーゼの変異体である、薬剤として使用するためのリパーゼ。
